There are 2 major variations that are often referred to as e-signatures. One is  electronic signatures and the other is digital signatures. For example this can take the form of a check box on a web page. If the User checks the box then s/he has agreed/signed to what the form (maybe T&C) says. On the other hand, Digital signatures are typically based on cryptography (ex: public key cryptography and certificates). In digital signatures, a secret (private key in case of PKI) is generated and associated to an individual that is authorized to sign. This secret (private key) has to be protected very well else someone can impersonate you. One of the key challenges in digital signatures is protecting this per user (could be device too) secret carefully (ex: private key is often stored in a smart card or crypto USB token). Unlike vanilla electronic signatures, Digital signatures can give you non-repudiation but it is still very hard to win in a court of law for various technical reasons.Another challenge with e-signatures is ensuring the client application (example: browser or PDF reader) is trust-able. Due to malware and MITB attacks this is easier said than done.Type of Authentication of the signer is another challenge.
