how're you guys I want to show you how to look at our data a little bit we have some sequences back and I put them on the public drive and also in Dropbox so I'm going to show you what we want to do is first we have four different sequence files for each plasmid so if I go to the public drive actually I'm not gonna go to the public drive because having a different place than you but if you guys go to the public drive then you should be able to see a folder here sequence data if you click on that open it up and you see all of these sequence files and there's some all different types of files and we won't have time to kind of get into a lot of what these are some of these allow you to go back and check the sequence quality and and throw out bad data points if you want what we're interested in is scrolling over or down to these dot seq files dot sequence files and those are the ones that were interested in and you'll notice that there's four of them for each plasmid and I named them with people's initials these are Colin's wands and then these are group ones Laura Diana and Shea and then it has low Meishan or cilantro or rabid abscess and then the PCR well that they originally came from and then the primers that were used to sequence them and so if you remember all these primers made allowed us to get overlapping sets of sequences sort of so what we have are our partial sequences that none of them individually covers the entire gap th insert in the plasmid but together if you could put them all together somehow they would cover that entire region and so that's I want to show you now is how to put them all together and get one continuous sequence so the first the way we're going to do that is using an online program called cap 3 and I'll show you how to find it all this if you google cap 3 it should be the first I don't know hundred thousand hits that come up is there's a lot of different servers that run the program NCBI hosts a version of it this one's very easy to use the very first one that comes up here on the do you APR a VI dot France site so I'm just going to go to that one and if you and you can see here it's got a field that lets you enter fast day files and so what we're going to do is we're going to enter all four of our fast day files for each plasmid that we're sequencing and this will assemble those sequences for us and so what you can do is you can go back and open up these files and you can see...
